Melanotan I: what it is and what the evidence says
A synthetic analog of melanocyte-stimulating hormone (alpha-MSH) that activates the MC1R receptor and increases eumelanin in the skin. It is the active ingredient of Scenesse (afamelanotide), approved for erythropoietic protoporphyria.
Last reviewed: August 16, 2026 · How we verify this
The ground it plays on
In aesthetics the route of administration decides almost everything. A peptide that works in a fibroblast culture may not cross intact skin, and then the formulation matters more than the active ingredient. That is why two products listing the same ingredient can give completely different results.
What it is studied for
Pigmentation, skin tone, light tolerance.
There is a distinction that almost always gets lost. A compound being studied for something is not the same as achieving it. And even where it works in a dish or in a mouse, that says little about whether it works in a person or whether the benefit is worth the risk. The section below therefore carries more weight than everything above it.
What the human evidence says
As afamelanotide it has two randomized, double-blind, placebo-controlled trials (N Engl J Med, 2015) showing more pain-free time in light and better quality of life in erythropoietic protoporphyria. In vitiligo there is one randomized trial combined with phototherapy and a phase 3 study whose results are expected in late 2026.
The limitation that matters most
The trials were run with the implant in adults with protoporphyria; aesthetic use of the lyophilized form has no trials of its own and no long follow-up. Its long-term effect on moles still needs study, and a 2024 cohort showed that on its own it does not correct the low vitamin D seen in these patients.
Regulatory status
Approved as Scenesse, an implant placed by a healthcare professional, for adults with erythropoietic protoporphyria by the EMA (2014), the FDA (2019), Australia (2020), and Canada (2026). It is not approved for vitiligo or for aesthetic use. It is not named on the 2026 WADA Prohibited List; athletes who compete should confirm with their federation.
Status can change and varies by country. Always verify the rules in force where you live before drawing conclusions.
Cautions and context
It can darken existing moles and freckles, so a mole map and periodic dermatology checks are advisable. Responses vary from person to person, and long-term safety data outside its approved indication are limited.
With a history of skin lesions or melanoma, dermatological assessment is essential.
What this page cannot give you
A reference page can take you this far. You know what Melanotan I is and where its research stands, which already puts you ahead of most readers. Four questions remain that no encyclopedia can answer for you, because the answers depend on your own situation:
- What would you measure beforehand, so that later you could tell whether anything actually changed?
- What would you watch along the way, and how consistently?
- What would you ask a professional, and in how much detail, so you do not walk out with a textbook answer?
- What would make you stop?
That is what the course is for
PeptiXtreme is ${CIF.LECCIONES} lessons that hand out no numbers. Each one ends with your starting point, what to watch along the way, the specific questions you take to a professional and the signals that mean stop. It includes per-module exams, a Certificate of Completion with a verifiable certificate ID, a Progress Tracker to follow how you are doing, and a human advisor you can actually write to.

Frequently asked questions
What is Melanotan I?
A synthetic analog of melanocyte-stimulating hormone (alpha-MSH) that activates the MC1R receptor and increases eumelanin in the skin. It is the active ingredient of Scenesse (afamelanotide), approved for erythropoietic protoporphyria. It is studied within the area of aesthetic.
Is there human evidence for Melanotan I?
As afamelanotide it has two randomized, double-blind, placebo-controlled trials (N Engl J Med, 2015) showing more pain-free time in light and better quality of life in erythropoietic protoporphyria. In vitiligo there is one randomized trial combined with phototherapy and a phase 3 study whose results are expected in late 2026.
Is it approved by any health agency?
Approved as Scenesse, an implant placed by a healthcare professional, for adults with erythropoietic protoporphyria by the EMA (2014), the FDA (2019), Australia (2020), and Canada (2026). It is not approved for vitiligo or for aesthetic use. It is not named on the 2026 WADA Prohibited List; athletes who compete should confirm with their federation.
What is the dose of Melanotan I?
We do not publish dosages. This is not an oversight. No single figure works for everyone: the regimen depends on a person's medical history, on whatever medication they already take, and on the goal they are chasing, which is something a professional weighs with the case in front of them. What you can build is the ability to walk into that appointment knowing what to ask.
Where can I learn to evaluate this properly?
This page tells you what Melanotan I is and what the evidence says. The PeptiXtreme course teaches the other half: what you would measure before considering anything, what you would watch, the specific questions you would take to a professional and when you would stop.
